Kajian Pengembangan Sistem Nanostructured Lipid Carriers (NLC) Pada Terapi Kanker Payudara

Abstract

Abstrak. Kanker payudara merupakan kanker dengan angka kejadian dan penyebab kematian tertinggi di Indonesia. Salah satu pengobatan kanker payudara adalah dengan kemoterapi menggunakan agen kemoterapi atau agen sitotoksik. Agen sitotoksik merupakan senyawa aktif yang dapat mengganggu pelekatan, mengubah morfologi, mempengaruhi laju pertumbuhan atau menyebabkan kematian pada sel. Maka dari itu dilakukan pengembangan sistem Nanostructured Lipid carriers (NLC) mengandung agen sitotoksik untuk meningkatkan aktivitas sitotoksik dari senyawa aktif pada terapi kanker payudara. Kajian dilakukan dengan Systematics Literature Review (SLR) terhadap artikel yang diperoleh pada database bereputasi yang memenuhi kriteria eksklusi dan inklusi. Hasil kajian menunjukan bahwa agen sitotoksik sudah banyak dikembangkan dalam sistem NLC diantaranya artemether dan docohexaenoic acid (DHA), dexibuprofen, docetaxel, doxorubicin, eucalyptol, imatinib, kurkumin, paclitaxel, quercetin, resveratrol dan tamoxifen. Formulasi sistem NLC menggunakan solid lipid dan likuid lipid yang distabilkan dengan penambahan surfaktan. Pengembangan sistem NLC terbukti dapat meningkatkan aktivitas sitotoksik senyawa aktif dilihat dari penurunan nilai IC50 yang dibandingkan dengan non NLC sebesar 1,25 hingga 24,94 kalinya.

Abstract. Breast cancer is a cancer with the highest incidence and cause of death in Indonesia. One of the treatments of breast cancer is chemotherapy using chemotherapeutic agents or cytotoxic agents. Cytotoxic agents are active compounds that can interfere with attachment, change morphology, affect growth rate or cause death in cells. Therefore, the development of a system of Nanostructured Lipid carriers (NLC) containing cytotoxic agents to increase the cytotoxic activity of active compounds in breast cancer therapy. The study was conducted with Systematics Literature Review (SLR) on articles obtained in reputable databases that meet exclusion and inclusion criteria. The results showed that cytotoxic agents have been widely developed in the NLC system including artemether and docohexaenoic acid (DHA), dexibuprofen, docetaxel, doxorubicin, eucalyptol, imatinib, curcumin, paclitaxel, quercetin, resveratrol and tamoxifen. NLC system formulations use solid lipids and liquid lipids stabilized by the addition of surfactants. The development of the NLC system is proven to increase the cytotoxic activity of active compounds seen from the decrease in IC50 value compared to non-NLC by 1.25 to 24.94 times.
