Uji Aktivitas Imunostimulan Ekstrak Etanol Buah Raspberry (Rubus idaeus) Berdasarkan Absorbansi Karbon pada Darah Mencit Swiss Webster

Abstract. The high prevalence of infectious diseases in Indonesia causes immunostimulant products to have a role in prevention as well as complementary therapy. One of the plants that has the potential as an immunostimulant is raspberry fruit. The purpose of this study was to determine the immunostimulant activity of ethanol extract of raspberry fruit based on carbon absorbance in the blood of mice using the carbon clearance method. Group 1 (negative control) was given CMC Na 0.5%, group 2 (positive control) was given Echinacae extract at a dose of 97.5 mg/kgBB and groups 3, 4, and 5 were given raspberry fruit ethanol extract at a dose of 100; 200 and 400 mg/kgBB. From the results of this study, it is known that there is a decrease in the average value of carbon absorbance in all test groups with a dose of 400 mg/kgBB having the highest value among the test groups, and based on statistical tests, there is a significant difference (p < 0.05) against the negative control group. These results indicate that the ethanol extract of raspberry fruit in doses of 100, 200, and 400 mg/kgBB has immunostimulatory activity, with the highest activity found at a dose of 400 mg/kgBB.
Abstrak. Tingginya prevelensi penyakit infeksi di Indonesia menyebabkan produk imunostimulan memiliki peran dalam pencegahan maupun sebagai terapi komplementer. Salah satu tanaman yang berpotensi sebagai imunostimulan adalah buah raspberry. Tujuan dari penelitian ini adalah untuk mengetahui aktivitas imunostimulan ekstrak etanol buah raspberry berdasarkan absorbansi karbon pada darah mencit menggunakan metode bersihan karbon. Pengujian dilakukan terhadap 25 ekor mencit yang dibagi menjadi 5 kelompok. Kelompok 1 (kontrol negatif) diberikan CMC Na 0,5%, kelompok 2 (kontrol positif) diberikan ekstrak Echinacae dosis 97,5 mg/kgBB, dan kelompok 3, 4, dan 5 diberikan ekstrak etanol buah raspberry dengan dosis 100; 200, dan 400 mg/kgBB. Ekstrak etanol buah raspberry diberikan secara peroral selama 7 hari dan di hari ke-8 ditentukan nilai absorbansi karbon pada darah tiap mencit. Dari hasil penelitian ini diketahui bahwa terdapat penurunan nilai rata-rata absorbansi karbon pada semua kelompok uji dengan dosis 400 mg/kgBB memiliki nilai yang paling tinggi di antara kelompok uji serta berdasarkan uji statistik terdapat perbedaan yang signifikan (p < 0,05) terhadap kelompok kontrol negatif. Hasil ini menunjukkan bahwa ekstrak etanol buah raspberry dosis 100, 200, dan 400 mg/kgBB memiliki aktivitas imunostimulan dengan aktivitas tertinggi terdapat pada dosis 400 mg/kgBB.
