Pengaruh Puasa Intermiten Kering terhadap Kadar Asam Urat Mencit (Mus musculus L.)

Lifestyle changes in the modern era such as changes in diet that more often consume high-fat foods cause various diseases, one of which is hyperuricemia which is caused by impaired excretion or excess production of uric acid. One of the non-pharmacological therapies that can be done to control uric acid levels is fasting. This study was conducted to determine the effect of dry intermittent fasting on uric acid levels in mice (Mus musculus L.). The method used in this study was a pure in vivo laboratory experiment with a completely randomized design. The subjects used were 28 adult male Swiss Webster mice divided into four groups. Group I or standard feed and did not do dry intermittent fasting, group II or high-fat feed and did not do dry intermittent fasting, group III or standard feed and dry intermittent fasting, and group IV or high-fat feed and dry intermittent fasting. The fasting period was carried out for 30 days with a fasting duration of 14 hours (5:00 PM-7.00 AM) and 10 hours of eating window. The results showed a significant increase in uric acid levels in the fasting group, namely groups III and IV compared to the non-fasting group (p <0.05). However, this increase is still within normal limits. During fasting, there is a change in metabolism from food to endogenous energy sources which ultimately causes an increase in uric acid levels due to the ketosis process. In addition, dehydration that occurs during fasting is a factor in increasing uric acid levels.
Abstrak. Perubahan gaya hidup di era modern seperti perubahan pola makan yang lebih sering mengonsumsi makanan tinggi lemak menyebabkan berbagai penyakit, salah satunya adalah hiperurisemia yang disebabkan karena adanya gangguan ekskresi atau kelebihan produksi asam urat. Salah satu terapi nonfarmakologi yang dapat dilakukan untuk mengendalikan kadar asam urat adalah dengan puasa. Penelitian ini dilakukan untuk mengetahui pengaruh puasa intermiten kering terhadap kadar asam urat pada mencit (Mus musculus L.). Metode yang digunakan dalam penelitian ini adalah eksperimental laboratorium murni in vivo dengan rancangan acak lengkap. Subjek yang digunakan adalah 28 mencit jantan dewasa galur Swiss Webster yang dibagi menjadi empat kelompok. Kelompok I atau pakan standar dan tidak melakukan puasa intermiten kering, kelompok II atau pakan tinggi lemak dan tidak puasa intermiten kering, kelompok III atau pakan standard dan puasa intermiten kering, serta kelompok IV atau pakan tinggi lemak dan puasa intermiten kering. Periode puasa dilakukan selama 30 hari dengan durasi puasa 14 jam puasa (17.00 – 07.00 WIB) dan 10 jam jendela makan. Hasil penelitian menunjukkan peningkatan signifikankadar asam urat pada kelompok puasa yaitu kelompok III dan IV dibandingkan kelompok tidak puasa (p <0,05). Namun peningkatan ini masih dalam batas normal. Pada saat puasa, terjadi perubahan metabolisme dari makanan ke sumber energi endogen yang menyebabkan pada akhirnya meningkatkan kadar asam urat karena adanya proses ketosis. Selain itu, dehidrasi yang terjadi pada saat puasa menjadi faktor peningkatan kadar asam urat.
