Uji Aktivitas Senyawa Aktif Kunyit (Curcuma Longa l.) sebagai Kandidat Antidepresan dengan Metode In Silico

Abstract

Abstract. A major depressive disorder is a depressive mood disorder. It can become a more serious condition if not handled appropriately and it can even lead to suicidal ideation. Therefore, appropriate treatment is needed. The standard antidepressant drugs that are used today have several side effects that may reduce patient compliance and thus indirectly reduce the effectiveness of treatment. As a result, new treatments are needed that can be as effective as current medications but have fewer side effects. One of the alternative medicine candidates is turmeric (Curcuma longa L.). This study aimed to test the activity of the active compound of turmeric (Curcuma longa L.) as an antidepressant candidate using the in silico method. This study was a descriptive observational study in the form of in silico analysis with a computational model using the applications PubChem, Swiss Target Prediction, and STRING to analyze the interaction between the active compounds contained in turmeric and target proteins that act as an antidepressants.The results of the search, selection, and analysis of turmeric's active compounds were found a number of 3 target proteins, namely ACHE, BACE1, and MAOA. The active compounds contained in turmeric that have the potential as antidepressants using the in silico method are toluene, 1-(4-hydroxy-3-methoxyphenyl) -7- (3,4-dihydroxyphenyl)-1,6-heptadiene -3,5-dione, curcumin. (curcumin I), demethoxycurcumin (curcumin II) and bisdemethoxycurcumin (curcumin III). Because of its performance in activating these three target proteins, turmeric has the potential to be used as an antidepressant.

Abstrak. Major depressive disorder merupakan gangguan mood depresi, yang jika tidak ditangani dengan baik dapat berkembang menjadi lebih berat dan berpotensi menimbulkan keinginan bunuh diri pada pasiennya. Oleh karena itu pengobatan yang tepat sangat dibutuhkan. Obat standar antidepresan yang dipakai saat ini diketahui memiliki beberapa efek samping yang kemungkinan dapat menurunkan kepatuhan pasien dan secara tidak langsung akan menurunkan pula efektifitas pengobatan. Oleh karena itu dibutuhkan adanya alternatif pengobatan yang memiliki efektifitas sama dengan obat standar namun memiliki efek samping yang minimal. Salah satu kandidat obat alternatif adalah kunyit (Curcuma longa L.). Penelitian ini bertujuan untuk melakukan uji aktivitas senyawa aktif kunyit (Curcuma longa L.) sebagai kandidat antidepresan dengan metode in silico.  Penelitian ini merupakan penelitian observasional deskriptif berupa analisis in silico dengan model komputasi menggunakan aplikasi PubChem, Swiss Target Prediction dan STRING untuk menganalisis interaksi antara senyawa aktif yang terkandung dalam kunyit dengan protein target yang berperan sebagai antidepresan. Hasil pencarian, seleksi, serta analisis berdasarkan ikatannya dengan senyawa aktif kunyit ditemukan sejumlah 3 protein target yang memiliki potensi antidepresan yaitu ACHE, BACE1 dan MAOA. Kandungan senyawa aktif kunyit yang berpotensi sebagai antidepresan dengan metode in silico adalah toluene, 1-(4-hydroxy-3-methoxyphenyl) -7- (3,4-dihydroxyphenyl)- 1,6 -heptadiene -3,5- dione, curcumin (curcumin I), demethoxycurcumin (curcumin II) dan bisdemethoxycurcumin (curcumin III). Kunyit memiliki potensi sebagai antidepresan melalui kerjanya dalam aktivasi dari ketiga protein target tersebut.
